Baclofen

Baclofen

It is derived from gama-aminobutyric acid (GABA) and is a highly effective antispastic with a spinal site of attack. Baclofen depresses monosynaptic and polysynaptic reflex transmissionwhich in turn inhibits the release of the excitatory amino acids glutamate and aspartate.The plasma elimination half-time of baclofen averages 3-4 hours. Its serum protein-binding rate is approx. 30 % and is eliminated largely in unchanged form.

Indications

Skeletal muscle spasticity of spinal and cerebral origin.


Dosage

Adults: 15 mg / day. Maintenance Dose: 30-75 mg / day in divided doses. Children : 0.75 -2 mg / kg / day in divided doses


Contra-Indications

Hypersensitivity to baclofen


Special Precautions

Pregnancy, epilepsy, psychiatric disorders, cerebrovascular or respiratory insufficiency, CNS depressants, antihypertensives, levodopa abrupt discontinuation. Paediatrics: Reduced dose necessary. Pregnancy: Safety not established. Lactation: Use with caution. Elderly: Reduced dose may be necessary due to adverse effect.


Side Effects

Sedation, drowsiness, nausea, respiratory depression,diminished cardiovascular functions


Drug Interactions

Drugs acting on CNS, alcohol, tricyclic antidepressants, levodopa with carbidopa.
