LOXAPINE

It is dibenzoxazepine which chemically resembles imipramine but has antipsychotic activity like chlorpromazine.

Indications

Manifestations of psychotic disorders. Acute chronic paranoid schizophrenia.


Dosage

Initial 10 mg twice daily. For severely disturbed : Initial upto 50 mg daily. Increase rapidly in 7-10 days. Usual therapeutic ose : 60-100 mg/day.


Contra-Indications

In comatose or severe drug induced depression states (alcohol, barbiturates, narcotics, etc.) in individuals with known hypersensitivity to benzodiazepines.


Special Precautions

In patients with history of convulsive disorders. CVD, glaucoma, tendency to urinary retention. Paediatrics: Not recommended. Pregnancy: Contraindicated. Lactation: Contraindicated. Elderly: Use with caution.


Side Effects

Insomnia, dizziness, tardiv dyskinesia, tachycardia, neuroleptics malignant syndrome, hypotension, nausea/vomiting, headache, paresthesia, polydypsia.


Drug Interactions

Additive CNS depression with sedatives, hypnotics, antihistamines and opiates. Orthostatic hypotension with MAOIs. Anticholinergic effects potentiated by antiparkinson agents & tricyclic antidepressants. Antagonises antihypertensive effect of guanethidine, methyldopa and clonidine.
