Triptorelin Acetate

Triptorelin acetate is a synthetic analogue of the naturally occurring gonadotropin-releasing hormone (GnRH). It is used as a depot preparation for sustained release in the body, reducing the frequency of dosing. It inhibits the release of gonadotropins, luteinizing hormone (LH), and follicle-stimulating hormone (FSH) through negative feedback, leading to decreased production of testosterone in males and estrogen in females.

Indications

Triptorelin acetate is indicated for: Treatment of advanced prostate cancer in men, reducing levels of testosterone which fuels tumor growth. Management of endometriosis in women, alleviating symptoms by reducing estrogen levels. Treatment of central precocious puberty (CPP) in children to delay early onset of puberty.


Dosage

Advanced prostate cancer: Typically, 3.75 mg is administered intramuscularly every 4 weeks. Endometriosis: A dose of 3.75 mg is typically given intramuscularly every 4 weeks for a duration of up to 6 months. Central precocious puberty (CPP): A dose of 22.5 mg is given every 3 months, with the specific dosing regimen adjusted based on individual response and monitoring.


Contra-Indications

Hypersensitivity to triptorelin, other GnRH analogues, or any other component of the formulation. Pregnancy and lactation: Triptorelin may cause harm to the fetus and should not be used during pregnancy or breastfeeding.


Special Precautions

Triptorelin should be used with caution in patients with pre-existing cardiovascular disease, as it can increase the risk of cardiovascular events. Bone density monitoring may be required, as long-term use can lead to osteoporosis. Close monitoring of children undergoing treatment for CPP is necessary to assess growth and development. Ensure adequate contraception during treatment, as triptorelin can cause temporary infertility.

Drug Interactions

Triptorelin may interact with other medications affecting hormone levels, such as hormone replacement therapy (HRT) or other GnRH analogues. Caution should be used when co-administering triptorelin with drugs known to prolong the QT interval, as the risk of arrhythmia may increase.
